Honoring giants in hematopoietic stem and progenitor cell biology

Imagine living in a time when there was no knowledge of hematopoietic stem and progenitor cells (HSPCs). Methods to assay HSPCs were lacking until pioneering transplantation studies of McCulloch and Till [1] and Ford et al. [2], published nearly 70 years ago, identified cells that could engraft and produce blood cells in irradiated mice. The field has rapidly exploded since then, and the development of technologies including flow cytometry [3] and methods to identify, isolate, and assay HSPCs have significantly improved our understanding of the different types of HSPCs, particularly mouse [4–6] and human [7,8].

Mixed lineage leukemia fusion oncoproteins often transform cells in particular developmental states without the need for additional mutations. By studying how this occurs, a wealth of mechanistic and biological insights have been gained. Here we reviewed some of the detailed molecular mechanisms leading to leukemia development as well as how understanding these mechanisms resulted in the development of targeted therapies, and ongoing efforts to anticipate and prevent resistance to targeted therapies.

ZC3H13 is highly expressed in acute myeloid leukemia (AML) and promotes leukemia progression by enhancing glycolysis, inhibiting apoptosis, and reducing oxidative stress. Mechanistically, ZC3H13 stabilizes FOXP1 expression via m6A methylation, driving metabolic reprogramming. These findings uncover a novel ZC3H13–FOXP1 axis and suggest potential therapeutic targets for AML treatment.

Transcription factor networks are the central hubs that control proper hematopoietic development. Their correct wiring is necessary to adjust the delicate balance between proliferation and differentiation and to ensure correct cellular function. A perturbation of these networks is a frequent hallmark of leukemia [1]. A puzzling observation is the high redundancy of transcription factor binding at the corresponding promoter and enhancer elements. Usually, a multitude of these proteins, often from the same or related protein families bind closely together at common sites.

The utilization of alteplase for the management of intracerebral thromboembolism is associated with an elevated risk of intracerebral hemorrhage. To mitigate this risk, we investigated the fibrinolytic effect of alteplase-loaded platelets as a preliminary step in the development of a biocompatible drug delivery system. In this context, the objective of this study was to ascertain whether alteplase could be loaded into platelets, identify any functional alterations in alteplase-loaded platelets, and determine whether alteplase-loaded platelets exert fibrinolytic effects.

Running a research laboratory is a multifactorial endeavor for junior and senior principal investigators (PIs). PIs are pivotal in overseeing research operations, securing funding, managing personnel with diverse skills, and driving scientific discoveries. Although most host institutions provide overarching research policies, several aspects of laboratory organization are often poorly defined. To streamline daily operations and interactions into an efficient and structured manner, PIs could significantly benefit by creating their own Laboratory Handbook.
